Bcl-2 Protein Family and Caspase Group Expression in SK-MEL-28 Cells Cultured in Vitiligo Patient¡¯s Serum

Abstract

Vitiligo is defined as an acquired, progressive disorder that selectively destroys melanocytes within the epidermis. There are numerous hypotheses about the etiology of vitiligo, but no data to definitively prove one theory over another. Apoptosis is a programmed cell death, and may be related to the removal of melanocytes. A family of highly specific cystein proteases known as caspase is responsible for the execution of the apoptosis. Apoptosis is regulated by the balance between the antiapototic and the proapoptotic proteins of the bcl-2 family.
This study was designed to investigate the effect of serum from vitiligo patients on the expression of caspase group and bcl-2 protein family in cultured melanocytes. Cultured SK-MEL-28 cells were treated with four viltiligo serums and analyzed for caspase-3, caspase-8, caspase-9 and bcl-2, bax, bad, bak expression using RT-PCR.
RT-PCR showed increased expression of caspase-3, caspase-8, caspase-9. In bcl-2 protein family, RT- PCR showed decreased expression of bcl-2 and increased expression of bax, bad, bak but, bax and bak were weakly increased.
Our results indicate that apoptosis of melanocyte in vitiligo is increased, but it is not clear that vitiligo is related to dysregulation of apoptosis regulatory molecules.
